CombiMatrix has created the first commercially available microarray stripping kit. The kit allows researchers to strip the target from a recently hybridized CustomArray™ enabling a subsequent re-hybridization. With the Stripping Kit, each CustomArray™ can be stripped and re-hybridized three times, yielding a total of four hybridizations and images per CustomArray™. The ability to re-use CustomArray™ microarrays reduces the cost per CustomArray™ to the researcher while eliminating problems associated with chip-to-chip reproducibility.

The stripping procedure is based on denaturation of DNA:DNA or DNA:RNA hybrids. This denaturation results in labeled targets being removed from the oligonucleotide probes and subsequently washed off the CustomArray™ while the original probes remain. The stripped CustomArray™ can then be re-hybridized with a new target sample and re-imaged.

CombiMatrix does not recommend increasing the number of re-uses beyond the recommended three (total of four hybridizations). Tests in our lab show that a CustomArray™ can be stripped up to three times without significantly decreasing the microarray quality.

The CustomArray™ Stripping Clamp enables the CustomArray™ to be stripped and re-hybridized up to three times. The Stripping Clamp is specially designed to withstand chemicals used for stripping and to provide appropriate pressure to prevent leakage from the stripping cap. The CustomArray™ Stripping Clamp is recommended for up to 100 stripping procedures.
